β-Secretase (BACE1) is a key enzyme involved in the production of β-amyloid peptides found in extracellular amyloid plaques of Alzheimer's disease (AD). This enzyme has been implicated to be an excellent target for anti-amyloid therapy of Alzheimerâs disease. The SensoLyte⢠β-Secretase Assay Kit provides a sensitive and efficient method for screening potential β-secretase inhibitors. This kit uses a β-secretase-cleavable FRET peptide substrate. In the intact peptide substrate, the fluorescence of the donor is quenched by the âdarkâ acceptor. The specific cleavage of the peptide by β-secretase physically separates the donor and acceptor, recovering the quenched fluorescence that is directly related to β-secretase activity.
